Under Pressure: How Pathogens Sense and Respond to Host-Induced Stresses

Pathogenic bacteria require dedicated mechanisms to adapt to the complex and fluctuating conditions they encounter within the host. One of the most universal adaptation mechanisms is the stringent response, which relies on the rapid accumulation of the alarmone nucleotides (p)ppGpp. Despite its central role, how the stringent response detects and integrates host-induced stresses remains elusive. In this seminar, I will present new insights into how infection-relevant stresses—such as iron limitation and oxidative damage—modulate the stringent response in major Gram-negative pathogens, including Salmonella enterica, Enterobacter cloacae, and Klebsiella pneumoniae. I will also discuss how (p)ppGpp reprograms bacterial transcription to promote stress adaptation and enhance virulence within the host. Together, these findings highlight a conserved mechanism by which pathogenic bacteria integrate metabolic stress into adaptive gene regulation, linking environmental sensing to transcriptional control during infection.
